I'm not new to the game, but would be to a league having only played solo in the past. Having looked around at a few league sites things seem to be pretty straightforward, but I wonder if anyone could give me a little more detail on the actual gameplay. Last year was my first without fantasy baseball, I didn't miss it at all, but I'm still looking for some way to fill that competitive void. Since I already spend enough time playing solo I wonder if a league might be something for me to look into.

What happens in pretty much every online league is that the commissioner runs every sim on his computer, then puts out the new league file for owners to download, and then export their changes. At the next sim, the commissioner imports what other owners have done since the last sim, and so on.
A season's length can vary greatly. In my league, for instance, sims happen thrice a week and cover 2 weeks' worth of in-game time, so this fast pace leads to a season being done in about 2 months (without the offseason). Other leagues have a slower pace, and/or sim less games at a time, so I suggest you look around for something that fits your schedule.
Online leagues are a great way to get competitive. Not only are your adversaries real humans, but that also means that it's harder to make lopsided trades.
